Teenage Mutant Ninja Turtles, Hitman: Agent 47) selaku sutradara dengan produsernya, Simon Kinberg (The New Mutants, X-Men Dark Phoenix), Lauren Shuler Donner (Constantine 2, Alpha Flight) dan Ryan Reynolds (The Hitman’s Bodyguard, Pokemon).
Tak hanya menjadi pemeran utama dan produser, Ryan Reynolds juga menulis skenario dari film keduanya tersebut berafiliasi dengan Rhett Reese (Life, Clue) dan Paul Wernick (Zombieland, Cowboy Ninja Viking). Dalam film ini, Deadpool akan membentuk sebuah tim bernama X-Force dengan berisikan Domino, Negasonic Teenage Warhead, Colossus, dan Bedlam. Mereka berusaha melindungi seorang mutan misterius yang diincar oleh Cable.
Film ini adalah sekuel atau kelanjutan dari film sebelumnya, Deadpool yang dirilis pada tahun 2016 dengan menceritakan awal mulanya seorang Wade Wilson menjadi Deadpool. Film Deadpool 2 menjadi film ke delapan dari X-Men series dimana Deadpool masuk kedalam X-Men universe. Film sebelumnya, Logan yang dirilis pada tahun 2017 kemudian kemudian film selanjutnya X-Men Dark Phoenix yang akan dirilis pada tahun 2019 bersama film The New Mutants.
Selaku rumah produksi, Marvel Entertainment, The Donners' Company dan Genre Films menghadirkan Josh Brolin (Avengers Infinity War, Hail Caesar!) sebagai villain dalam film ini berjulukan Nathan Summers atau yang dikenal sebagai Cable. Turut serta pemain lain menyerupai Zazie Beetz (Slice, High Flying Bird), Stefan Kapicici (Underdogs Rising, Hitac), Bill Skarsgard (IT, Villains), Brianna Hildebrand (The Voice Inside, Tragedy Girls), T. J. Miller (Ready Player One, Underwater) dan Morena Baccarin (DeLorean, Ode to Joy).
Film Deadpool 2 telah diumumkan 3 hari sebelum perilisan film Deadpool (2016) dan untuk sekuelnya, Deadpool 3 sudah dalam proses pengembangan.
Selaku pihak pendistributor, 20th Century Fox menjadwalkan film Deadpool 2 akan tayang pada pertengahan bulan Mei dengan tagline-nya "Prepare for the second coming". Diberitahukan kalau film Deadpool 2 akan berating usia untuk Dewasa atau 17 tahun keatas menyerupai pada film pertama.
Sebelumnya, Film Deadpool 2 dijadwalkan rilis pada bulan 1 Juni 2018, namun berubah sesudah film Avengers Infinity War memajukan aktivitas penayangannya.

Pada bulan Oktober 2016, Tim Miller memutuskan untuk tidak menyutradarai lagi sekuel film Deadpool sebab mempunyai problem dengan sang pemain drama utama, Ryan Reynolds selama masa pra-produksi.
Ditahun 2018, Josh Brolin akan bermain sebagai dua karaktor Marvel Comics. Keduanya yaitu villain dimana ia akan menjadi Thanos dalam film Avengers Infinity War yang dirilis bulan April 2018 dan Cable dalam film Deadpool 2 yang dirilis sebulan setelahnya, Mei 2018.
Film Deadpool bersama dengan The New Mutants (2019) dan X-Men Dark Phoenix (2019) mengambil latar waktu yang sama dimana bencana dalam film The New Mutants mengambil lokasi di Boston, Massachusetts, Amerika Serikat, Deadpool 2 di Vancouver, British Columbia, Canada dan X-Men Dark Phoenix di Montreal, Quebec, Kanada.
Black Tom yaitu mutan yang sanggup memanipulasi, mengikat dan memproyeksikan energi melalui kehidupan tanaman. Dia juga sanggup mengeluarkan ledakan dengan objek kayu, biasanya berupa Shillelagh, tongkat kayu dengan tombol besar dibagian atas.
